阿爾茨海默病(Alzheimer's disease,AD)是老年人中的主要癡呆類型。目前臨床用藥物只能緩解癥狀、無治療效果,因此,亟需開發有效的AD治療藥物。本研究以三轉基因AD模型小鼠3×Tg-AD為研究對象,對2月齡AD小鼠分別給予0.2和1.0 mmol/L乙基麥芽酚氧釩(BEOV)飲用水2個月,采用曠場實驗、高架十字迷宮、Y迷宮3種行為學測試法,發現BEOV可以顯著改善4月齡AD小鼠的自發活動、探索和記憶能力、減輕小鼠的焦慮狀態。采用活體熒光成像技術在體追蹤檢測2.5和3.5月齡小鼠腦皮層神經元樹突棘的變化,發現YFP熒光小鼠樹突棘數量呈增多趨勢;而3×Tg-AD與YFP小鼠雜交后代AD-YFP小鼠的樹突棘則呈下降趨勢。與AD-YFP鼠相比較,補充1.0 mmol/L BEOV的AD-YFP小鼠,腦皮層神經元總樹突棘、蘑菇型樹突棘、細小型樹突棘數量均呈顯著增加,說明BEOV可保護神經元、減少小鼠樹突棘的丟失。采用電感耦合等離子體質譜(ICP-MS)檢測3×Tg-AD鼠腦金屬離子水平,結果表明,1.0 mmol/L BEOV能顯著提高鼠腦V和Se的水平、降低Fe、Zn、Hg、Pb、Bi和Ni的水平,說明V可與Se協同調控AD鼠腦中多種金屬離子水平。本研究表明,BEOV可通過調控AD鼠腦中多種金屬離子的內穩態平衡,保護神經細胞,抑制AD小鼠神經元樹突棘丟失,從而增強小鼠記憶能力,干預AD病理進程。
